Okay, now let's start from the beginning:

- I'd like to see your current plustek.conf file
- perform sane-find-scanner as normal user
- login as root and do:
* sane-find-scanner -v
* SANE DEBUG PLUSTEK=12 ; scanimage -L
- as normal user do:
* SANE DEBUG PLUSTEK=12 ; scanimage -L

- send me the output of 
lsmod

Please no reboot between the tests!
And send it either to the mailing-list or directly to me but not both...
